PREPARATION OF PROTEIN FOR MICRO- SEQUENCING:

Run sample in acrylamide-SDS gel.

Electroblot onto Immobilon PSQ transfer membrane, using Tris-glycine, methanol at 5 C at 200 mA overnight.

Stain filter for 5-10 minutes with Coomasie.

Destain with 50% methanol, 10% acetic acid.

Cut the desired band.

Note: Make sure to have marker in the gel.

ELECTROBLOTTING SOLUTION:

25 mM Tris-Base.

190 mM glycine.

20% methanol.
