Lab Vectors
List of vectors:
pT7KO1c - for replacing the first allelle of
glutamate
dehydrogenase of T. brucei with T7 RNA polymerase. Click
here (shift-click if using Netscape on
a PC, Ctl-click if using Netscape on a Mac)) to download a file with the
features that can be viewed with the NTI viewer. Click
here to download the
NTI Viewer for any platform. Click here to see this sequence in Genbank
text format.

pEKA2 revised - for replacing the first allelle of HGPRT in L.
tarentolae with T7 RNA polymerase. Click here to download a file to view with the NTI
Viewer. Click here to see a text file in
Genbank Format.

pHD360 (from C. Clayton). Click here to
download a file to view with the NTI Viewer. Click here to see a text file in GenBank format.

pLEW79 corrected (Wirtz
and Cross). Click here to see a text
file in GenBank format.

pX (Beverley).

8. pTS0-Hyg (C.C.
Wang). Click here to see the file in Genbank
format.

pZJM plasmid for RNAi in T. brucei (Englund
lab). Click here to see the file in Genbank format.

pTUTi-H2H. This is the construct we made for cloning Tb TUTase
fragments in a head to head configuration for RNAi. Click
here to see sequence in Genbank format.

pSAP1. This is a new dual epitope TAP vector with the strepavidin
binding peptide (Wilson
et al, 2001) substituted for the calmodulin binding peptide. Click
here for the sequence in Genbank format. If
you have the
free NTI viewer, it will open in graphic form. The
plasmid has the L. major RET2 gene in the expression site. Release this
fragment with BamHI and XbaI and clone your gene of interest in frame
with the C-terminal tags. Select in E. coli with 50 ug/ml amp.

pXLtREL1-mod. This is the original TAP vector with
the calmodulin binding peptide and the protein A peptide (Aphasizhev
et al., 2002). Remove the insert with XbaI and BamHI and clone in
your gene. Click here to see the sequence
in Genbank format. If you have the free NTI viewer, it will open in
graphic form. Otherwise, download and open in a text editor such as
Notepad.
